Direct identification of microorganisms from thioglycolate broth by MALDI-TOF MS.

We developed an easy MALDI-TOF MS-based assay to identify microorganisms directly from thioglycolate broth. A total of 101 positive thioglycolate broths inoculated with 15 different kinds of samples were evaluated. In 91 samples (90.1%), direct MALDI-TOF MS identifications were the same as those obtained after conventional laboratory procedures including subcultures. In 10 samples misidentified by direct processing, yeasts or mixed cultures grew in the thioglycolate subcultures, or high cellular debris hampered a correct analysis. Monitoring pH and ORP in a SHARON reactor

This paper analyses the valuable information provided by the on-line measurements of pH and oxidation reduction potential (ORP) in a continuous single high ammonia removal over nitrite (SHARON) reactor. A laboratory-scale SHARON reactor equipped with pH, ORP, electric conductivity and dissolved oxygen (DO) probes has been operated for more than one year. Nitrogen removal over nitrite has been achieved by adding methanol at the beginning of anoxic stages. Die Depolymerisation von Trioxan mit verschiedenen Phosphorsauren wurde im Temperaturbereich von 100–160°C untersucht. Sie fuhrt zu einem temperaturabhangigen Gleichgewicht zwischen Trioxan und Formaldehyd, das oberhalb 180°C weitgehend auf der Seite des Formaldehyds liegt. Zu einer schnellen Einstellung des Gleichgewichtes eignen sich Ortho- und Pyrophosphorsaure. Die bevorzugte Nebenreaktion ist die Bildung von Methylformiat; die Ausbeute an Methylformiat wird durch Erhohung der Reaktionszeit und der Katalysatoroberflache vergrosert. Seeking the Holy Grail: robust chronologies from archaeology and radiocarbon dating combined

The strengths of formal Bayesian chronological modelling are restated, combining as it does knowledge of the archaeology with the radiocarbon dating of carefully chosen samples of known taphonomy in association with diagnostic material culture. The risks of dating bone samples are reviewed, along with a brief history of the development of approaches to the radiocarbon dating of bone. In reply to Strien (2017), selected topics concerned with the emergence and aftermath of the LBK are discussed, as well as the early Vinča, Ražište and Hinkelstein sequences. The need for rigour in an approach which combines archaeology and radiocarbon dating is underlined.

The effect of pH and nitrite concentration on the activity of the nitrite oxidizing bacteria (NOB) in an activated sludge reactor has been determined by means of laboratory batch experiments based on respirometric techniques. The bacterial activity was measured at different pH and at different total nitrite concentrations (TNO 2). The experimental results showed that the nitrite oxidation rate (NOR) depends on the TNO 2 concentration independently of the free nitrous acid (FNA) concentration, so FNA cannot be considered as the real substrate for NOB.
